چکیده
Despite the fact that photodynamic therapy (PDT) has promising clinical prospects, its therapeutic efficacy is still restricted by hypoxic tumor microenvironment (TME). Therefore, how to suitably employ hypoxia vital for optimizing PDT. Herein we developed a hypoxia-activated prodrug combination with A hypoxia-responsive camptothecin (CPT)-based and near-infrared (NIR) BODIPY photosensitizer were coloaded within ?-cyclodextrin-based polymer through supramolecular host–guest recognition obtain nanoparticles named CPT-BODIPY-PNs. After being internalized tumor, oxygen in tissues was rapidly consumed moiety generate reactive species under NIR irradiation. The efficiently activated this intensified condition release chemotherapeutic CPT drug. This system makes best use of TME achieve effectively combined PDT chemotherapy, offering an approach cancer treatment.

Collaboration between hepatic and intratumoral prodrug activation in a P450 prodrug-activation gene therapy model for cancer treatment.
Presently, we investigate the mechanisms whereby intratumoral expression of a cyclophosphamide-activating hepatic cytochrome P450 gene enhances therapeutic activity when cyclophosphamide is given on an every 6-day (metronomic) schedule. In P450-deficient 9L gliosarcomas grown in severe combined immunodeficient mice, metronomic cyclophosphamide substantially decreased tumor microvessel density a...
متن کاملA mitochondrial-targeted prodrug for NIR imaging guided and synergetic NIR photodynamic-chemo cancer therapy† †Electronic supplementary information (ESI) available. See DOI: 10.1039/c7sc03454g
Nontoxic prodrugs, especially activated by tumor microenvironment, are urgently required for reducing the side effects of cancer therapy. And combination of chemo-photodynamic therapy prodrugs show effectively synergetic therapeutic efficiency, however, this goal has not been achieved in a single molecule. In this work, we developed a mitochondrial-targeted prodrug PNPS for near infrared (NIR) ...
متن کاملEffect of POR knockout on hypoxic prodrug activation
Background: NADPH:cytochrome P450 oxidoreductase (POR) catalyses one-electron reduction of hypoxia-activated prodrugs (HAP). Results: Knockout of POR in tumour cells inhibited activation of some HAP (e.g. 5nitroquinoline SN24349) but not others (e.g. PR104A, TH-302). Conclusion: POR at endogenous levels plays a variable role in HAP activation. Significance: POR expression alone is not a suffici...
متن کاملStrategies for enzyme/prodrug cancer therapy.
The selective activation of prodrug(s) in tumor tissues by exogenous enzyme(s) for cancer therapy can be accomplished by several ways, including gene-directed enzyme prodrug therapy (GDEPT), virus-directed enzyme prodrug therapy (VDEPT), and antibody-directed enzyme prodrug therapy (ADEPT). The central part of enzyme/prodrug cancer therapy is to deliver drug-activating enzyme gene or functional...
متن کاملTargeting hypoxic microenvironment of pancreatic xenografts with the hypoxia-activated prodrug TH-302
Previous reports have suggested that the hypoxic microenvironment provides a niche that supports tumor stem cells, and that this might explain clinical observations linking hypoxia to metastasis. To test this, we examined the effects of a hypoxia-activated prodrug, TH-302, on the tumor-initiating cell (TIC) frequency of patient-derived pancreatic xenografts (PDX).The frequencies of TIC, measure...
